要旨本文 | MicroRNAs (miRNAs) play significant roles in regulatory mechanisms in mycorrhizal symbiosis. In the previous study, we identified the differentially expressed miRNAs (DE-miRNA) in roots of Populus tomentosa inoculated by Cenococcum geophilum by miR-seq. In this research, we selected four DE-miRNAs (miR164c, miR172a, miR319a and miR396a) as candidate miRNAs to reveal how miRNAs regulate ectomycorrhizal formation. We constructed the overexpressed, knocked-down and knocked-out transgenic mutants of P. tomemtosa for these four DE-miRNAs. At present, the overexpressed and knocked-down transgenic mutant lines were successfully regenerated. The knocked-out mutants of P. tomentosa by CRISPR/Cas9 system are being prepared. As the next step, we will investigate the regulating mechanisms of four DE-miRNAs by using the constructed transgenic mutants. |
